The CD34 MicroBead Kit, formerly termed Direct CD34 Progenitor Cell Isolation Kit, is a single-step labeling system which allows the fast and easy isolation of CD34+ cells. The CD34 MicroBead Kit contains MicroBeads conjugated to the monoclonal mouse anti-human CD34 antibody QBEND/10 and FcR Blocking Reagent to avoid unspecific labeling of cells via Fc receptors. By using direct MicroBeads instead of an indirect labeling system, the washing step during the labeling procedure is abolished and the resulting potential loss of cells is avoided. CD34+ cells can be isolated from peripheral blood, bone marrow, leukapheresis product, or cord blood. The CD34 antigen, a single transmembrane glycoprotein, is expressed on a heterogeneous population of hematopoietic stem and progenitor cells that display a wide spectrum of proliferative capacity and lineage commitment. Furthermore, CD34 expression is also found on hemangioblasts3, endothelial progenitor cells4–6, and mature endothelial cells5. CD34 was also shown to be expressed by embryonic stem cell lines during their differentiation to hematopoietic and endothelial lineages.7 A large-scale CD34 MicroBead Kit is available for experiments involving large numbers of cells.
