IVT templates for the generation of in situ probes
Easy preparation of highly specific RNA probes!

In situ hybridization (ISH) and northern blot analysis are common and powerful tools for the detection and quantification of mRNA. To achieve reliable and reproducible results, it is essential to use single-stranded RNA probes that have a high sequence specificity.
Often, the generation of these high-quality RNA probes turns out to be more tedious and time-consuming than expected.
Miltenyi Biotec's sequence-verified EasyProbe Transcription Templates are ready-to-use, linear DNA templates (in vitro transcription templates) for the generation of specific RNA probes for in situ hybridization or northern blot analysis.

Principle
EasyProbe Transcription Templates are linear, double-stranded DNA templates ready-to-use for in vitro transcription (IVT). Delivered as lyophilizates, the template only needs to be solubilized in water before IVT (fig. 1). Depending on the individual experimental setting, the RNA probes can be generated using diverse labels such as fluorescence (e.g. Cy5 and Cy3), digoxygenine (DIG), biotin, or radioactive isotopes.

The gene-specific segment of the templates ranges from 250–400 basepair (bp). The supplied 200 ng lyophilizate will suffice for five IVT reactions producing enough RNA probes for 20–50 in situ hybridizations experiments or northern blot analyses.
