Anti-Sca-1
Description
Clone D7 specifically recognizes the mouse Sca-1 antigen. Sca-1 (stem cell antigen-1) is an 18 kDa GPI-linked surface protein of the Ly-6 family (Ly-6A/E). The Anti-Sca-1 antibody recognizes both Ly-6E.1 and Ly-6A.2, which are gene products of two Ly-6A/E alleles expressed in different mouse strains (Ly-6E.1: e.g. BALB/c, C3H, NZB; Ly6A.2: e.g. C57BL/6, SJL, 129, AKR).
Sca-1 is expressed on stem cells in bone marrow with hematopoietic differentiation potential. Sca-1 is also expressed on a population of CD105+ hematopoietic stem cells (HSCs). This population has a long-term repopulating (LTR) capacity and is therefore termed LTR-HSCs1,2. Sca-1+ cells from bone marrow also possess nonhematopoietic differentiation potential as they are able to give rise to hepatocytes in vivo3 and neural cells in vitro4.
Sca-1 expression is also found on distinct subpopulations of bone marrow and peripheral B cells, myeloid cells, thymic and peripheral lymphocytes, and early intrathymic T cell precursors.
Furthermore, Sca-1 is expressed on stem cells in a variety of nonhematopoietic tissues, such as adult liver5, heart6, and prostate7.
Applications
The Anti-Sca-1 antibody can be used to identify and characterize Sca-1+ cells by flow cytometry or fluorescence microscopy. This antibody is suitable for MACS® Control applications. For further details, please refer to the data sheet of the separation reagent.

Mouse bone marrow cells were stained with Anti-Sca-1 antibodies conjugated to FITC (A), PE (B), APC (C), or biotin (D) and analyzed by flow cytometry. Cells stained with Anti-Sca-1-Biotin were additionally stained with Anti-Biotin-APC.
