| Description |
Clone JF05-1C2.4.1 specifically recognizes the mouse plasmacytoid dendritic cell antigen-1 (mPDCA-1). mPDCA-1 is specifically expressed on mouse plasmacytoid dendritic cells (PDCs) — a subset of CD11c+ dendritic cells detected at low frequency in all lymphoid tissues, peripheral blood, and some non-lymphoid tissues. In mouse spleen, bone marrow, and lymph nodes, mPDCA-1 is exclusively expressed on cells which are CD11clow, Ly-6Chigh, CD45R (B220)+, MHC Class IIlow, CD8alow/–, CD80low/–, CD86–, CD40–, CD11b–, CD90–, CD49b (DX5)–, CD3–, CD19–, i.e. on cells with the phenotype of mouse PDCs. Multi-color fluorescent staining of spleen cells clearly revealed that all CD11clow Ly-6Chigh CD45R (B220)+ PDCs are mPDCA-1+ and that mPDCA-1 expression is restricted to PDCs. |
| Applications |
| Anti-mPDCA-1 antibodies allow the direct, single-color detection of mouse PDCs by flow cytometry, fluorescence microscopy, or immunohistochemistry. They are an excellent tool for the functional analysis of PDCs directly isolated from tissues, in vitro-generated from bone marrow cell cultures, or by in vivo depletion. Furthermore, Anti-mPDCA-1 antibodies are suitable for studies on PDCs in experimental mouse models of infection or autoimmune disease regarding tissue distribution, frequency, specific localization within lymphoid organs, and recruitment to inflamed tissues. The antibodies are also recommended for evaluation of the purity of mouse PDCs isolated with the Plasmacytoid Dendritic Cell Isolation Kit II, Anti-mPDCA-1 MicroBeads or CD11c MicroBeads. |

Identification of CD11c+ Ly-6C+ CD45R (B220)+ PDCs with Anti-mPDCA-1. Splenocytes isolated from BALB/c mice were analyzed by multi-color fluorescent staining with CD11c, Anti-Ly-6C, CD45R (B220), and Anti-mPDCA-1. Cells expressing Ly-6Chigh CD45R (B220)high (A) are also positive for CD11c and mPDCA-1 (B). Note that all CD11clow Ly-6Chigh CD45R (B220)+ PDCs are mPDCA-1+ and that mPDCA-1 expression is restricted to PDCs only. |
