Anti-IL-17
Description
Clone CZ8-23G1 detects human interleukin 17. IL-17 (IL-17A, CTLA8) is produced by Th17 cells, a T helper subset distinct from Th1 and Th2 cells. These cells also secrete IL-17F and IL-22 and express the NK cell marker CD1611. IL-17 secretion has also been described for other cell types, for example, CD8+ memory T cells.
Th17 cells are involved in the recruitment of neutrophils to control early stages of infections by a number of pathogens, such as extracellular bacteria and fungi. IL-17 and Th17 cells play an important role in immune-mediated inflammatory diseases, such as rheumatoid arthritis, psoriasis, multiple sclerosis, asthma, and inflammatory bowel disease.
Anti-IL-17 antibodies allow intracellular staining of IL-17.
Applications
Identification and enumeration of IL-17–producing cells by flow cytometry or fluorescence microscopy or antigen-specific T cells upon restimulation with the respective antigen.
Analysis of cytokine production of rare cells by in-column staining technology, in combination with magnetic enrichment of cells
Cross-reactivity
  • Rhesus monkey (Macaca mulatta)
  • Cynomolgus monkey (Macaca fascicularis)

Figure 1
Human PBMCs were incubated with or without CytoStim for 6 hours. After 2 hours brefeldin A was added. The cells were harvested, fixed, permeabilized, and intracellularly stained with Anti-IL-17 antibodies conjugated to FITC (A), PE (B), or APC (C). Cell surface staining was performed with CD4-FITC or CD4-PE and CD154-PE or CD154-APC. Cells were analyzed using the MACSQuant® Analyzer. Gating was performed according to CD4 expression and side scatter properties of the cells. Cell debris was excluded from the analysis in a propidium iodide versus Anti-IL-17-PE dot plot.
