ErbB-2 Tumor Cell Enrichment and Detection Kit
Description
The ErbB-2 Tumor Cell Enrichment and Detection Kit has been developed for the highly sensitive detection of epithelial tumor cells in non-epithelial tissues, for example, peripheral blood or bone marrow. Tumor cells are specifically enriched based on the expression of the ErbB-2 antigen, which is also known as HER2/neu. Subsequently, the enriched cells are subjected to immunocytochemical staining based on the expression of cytokeratins. First, cells are labeled with Anti-ErbB-2 MicroBeads and positively selected on a first separation column. Then, ErbB-2+ cells are fixed, immobilized within a second MACS® Column, and subjected to intracellular staining of cytokeratin filaments.
The detection reagent is a cocktail of two alkaline phosphatase-conjugated cytokeratin-specific antibody clones, CK3-11D5 and CK3-3E4. After elution from the column, cells are transferred onto a microscope slide, incubated with the substrate, and are then ready for analysis by light microscopy. During the entire staining procedure, the cells remain bound to the column matrix, thus minimizing cell loss.
Applications
Enrichment and detection of disseminated carcinoma cells from blood, bone marrow and lymphoid tissue.

100 cells of the tumor cell line SK-BR3 were spiked into 5Ă—107 PBMCs of a normal donor. Thereafter, the tumor cells were enriched and analyzed using the ErbB-2 Tumor Cell Enrichment and Detection Kit. The picture shows an enriched ErbB-2+ tumor cell.
