CD326 (EpCAM) Tumor Cell Enrichment and Detection Kit

Description

The CD326 (EpCAM) Tumor Cell Enrichment and Detection Kit has been developed for the highly sensitive detection of epithelial tumor cells in non-epithelial tissue, for example, peripheral blood or bone marrow.
With the CD326 (EpCAM) Tumor Cell Enrichment and Detection Kit, tumor cells are specifically enriched by targeting their expression of CD326, also known as human epithelial antigen (HEA), epithelial cell adhesion molecule (EpCAM), or epithelial-specific antigen (ESA). Subsequently, enriched cells are immunocytochemically stained for their cytokeratin expression using the in-column staining technology. First, cells are labeled with CD326 (EpCAM) MicroBeads and enriched using MACS^® Technology. Secondly, CD326⁺ cells are fixed, immobilized within a second MACS^® Column, and cytokeratin filaments intracellularly stained (see also product Inside Stain Kit). The detection reagent is a cocktail of two alkaline phosphatase–conjugated cytokeratin-specific antibodies, clones CK3-11D5 and CK3-3E4. After elution from the column, cells are transferred onto a microscope slide, incubated with the substrate, and then analyzed by light microscopy. During the entire staining procedure, the cells remain bound to the column matrix, thus minimizing cell loss.

Enrichment and detection of disseminated carcinoma cells from blood, bone marrow and lymphoid tissue.

Cells of the tumor cell line BT474 were spiked into 5×10⁷ PBMCs of a healthy donor. Thereafter, the tumor cells were enriched and analyzed using the CD326 (EpCAM) Tumor Cell Enrichment and Detection Kit.
