The CD8+CD57+ T Cell Isolation Kit was developed for the isolation of CD57+ cytotoxic T cells. The separation of CD8+CD57+ T cells is performed in a two-step procedure. In a first step, CD8+ cells are magnetically labeled with CD8 MultiSort MicroBeads and isolated by positive selection. After the release of the magnetic particle, the CD8+CD57+ target cells are magnetically labeled with CD57 MicroBeads to be positively selected a second time. The CD57 antigen has been shown to be expressed on late-stage CD8+ effector cells.1 CD57 expression is also increased on proliferation-incompentent or chronically activated CD8+ T cells in persistent viral infections, such as HIV.2 |

| CD8+CD57+ cells were separated from human PBMCs using the CD8+CD57+ T Cell Isolation Kit and an LS Column in a MidiMACS™ Separator for the first positive selection, followed by the separation over an MS Column in a MiniMACS™ Separator. Cells were fluorescently stained with CD8-FITC and CD57-PE. |
