Background information The CD8+CD45RA+ Effector T Cell Isolation Kit is ideal for the isolation of cytotoxic T cells that express high levels of perforin and granzyme B1. Isolated cells can be analyzed for interactions with other cell types and degranulation or secretion of effector molecules. Detailed separation procedure The isolation of CD8+CD45RA+ effector T cells is performed in a two-step procedure. First, non-target cells are indirectly magnetically labeled for depletion with biotin-conjugated antibodies against CD45RO and CD56 and an APC-conjugated antibody against CD197 (CCR7), as well as Anti-APC MicroBeads and Anti-Biotin MicroBeads. In the second step, the CD8+CD45RA+ effector T cells are directly labeled with CD8 MicroBeads and isolated by positive selection from the pre-enriched cell fraction. |
