Detailed separation procedure Non-target cells are indirectly magnetically labeled by using a cocktail of biotin-conjugated antibodies against CD4, CD11c, CD14, CD15, CD16, CD19, CD34, CD36, CD45RA, CD56, CD57, CD61, CD123, CD141, TCR γ/δ, and CD235a (glycophorin A), and Anti-Biotin MicroBeads. Isolation of highly pure CD8+ memory T cells is achieved by depletion of the magnetically labeled cells.
Downstream applications CD8+ memory T cells isolated with the CD8+ Memory T Cell Isolation Kit are used when direct labeling of cell surface molecules could interfere with downstream applications, e.g., in studies on memory T cell activation. Untouched CD8+ memory T cells can also be used for phenotypical and functional studies. |
