Background information CLA (cutaneous lymphocyte-associated antigen) is an inducible carbohydrate modification of P-selectin glycoprotein ligand 1 (PSGL-1) that functions as a skin-homing receptor. CLA is found on a minor subset of human T cells that infiltrate the skin. The ligand of CLA, E-selectin, is expressed on vascular endothelium. In peripheral blood, CLA is not only expressed on CD4+ memory/effector T cells but also on subsets of CD8+ T cells, B cells, NK cells, dendritic cells, and monocytes. Detailed separation procedure First, non-CD4+ T cells are first magnetically labeled and depleted using a cocktail of biotin-conjugated antibodies and Anti-Biotin MicroBeads. Thereafter, the CD4+CLA+ T cells are indirectly magnetically labeled with Anti-CLA-PE and Anti-PE MicroBeads to be isolated by positive selection. Downstream applications Specific isolation of CD4+CLA+ skin-homing memory/effector T cells from peripheral blood for phenotypical and functional characterization. |

| CD4+CLA+ T cells were isolated from PBMCs using the CD4+CLA+ T Cell Isolation Kit, an LS Column and a MidiMACS™ Separator for enrichment of CD4+ T cells, and an MS Column and a MiniMACS™ Separator for subsequent positive selection of CD4+CLA+ T cells. Cell debris and dead cells were excluded from analysis based on scatter signals and PI fluorescence. |
