Anti-CLA MicroBead Kit
Description
The Anti-CLA MicroBead Kit was developed for the positive selection or depletion of human CLA+ cells from PBMCs, lymphoid and non-lymphoid tissue for phenotypical and functional characterization.
Details
Background information
The cutaneous lymphocyte-associated antigen (CLA) is an inducible carbohydrate modification of P-selectin glycoprotein ligand 1 (PSGL-1) and functions as ligand for E-selectin. CLA is a skin-homing receptor and is found on a minor subset of human T cells that infiltrate the skin. This post-translational modification of PSGL-1 is discussed to serve as mechanism to regulate homing of CD4+ and CD8+ memory/effector T cells from peripheral blood to the skin; an immune response that plays an essential role during many inflammatory and certain malignant skin diseases.
In peripheral blood, CLA is expressed on skin-homing memory/effector T cells, on few B cells, NK cells, and almost all monocytes and dendritic cells, including Langerhans cells in the skin.
The Anti-CLA MicroBead Kit can be used in combination with MACS® Cell Isolation Kits or MACS MultiSort Kits, for example, with the CD8+ T Cell Isolation Kit for the isolation of CD8+CLA+ memory/effector T cells.

Detailed separation procedure
The isolation is performed by a two-step procedure. The CLA+ cells are fluorescently and indirectly magnetically labeled with Anti-CLA-PE and Anti-PE MicroBeads.
Columns
For positive selection: MS, LS, XS, or autoMACS Columns. For depletion: LD, CS, D, or autoMACS Columns.
