Description
The CD4+ T Cell Isolation Kit II is an indirect magnetic labeling system for the isolation of untouched CD4+ T helper cells from human PBMCs. Non-CD4+ T cells, i.e., CD8+ T cells, γ/δ T cells, B cells, NK cells, dendritic cells, monocytes, granulocytes, and erythroid cells, are indirectly magnetically labeled by using a cocktail of biotin-conjugated antibodies against CD8, CD14, CD16, CD19, CD36, CD56, CD123, TCR γ/δ, and CD235a (glycophorin A). These cells are subsequently magnetically labeled with Anti-Biotin MicroBeads for depletion. Thereby, highly pure CD4+ T cells are obtained.
Applications
T helper cells isolated with the CD4+ T Cell Isolation Kit II can be used when direct labeling of CD4+ T cell surface molecules could interfere with downstream applications, e.g., in studies on T helper cell activation.1–5 Untouched CD4+ T cells may also be used for the isolation of specific T helper cell subsets, such as naive6 or memory CD4+ T cells and activated CD4+ T cells.
