Detailed separation procedure Non-CD4+ T cells, i.e., CD8+ T cells, γ/δ T cells, B cells, NK cells, dendritic cells, monocytes, granulocytes, and erythroid cells, are labeled by using a cocktail of biotin-conjugated antibodies against CD8, CD14, CD16, CD19, CD36, CD56, CD123, TCR γ/δ, and CD235a (glycophorin A). These cells are subsequently magnetically labeled with Anti-Biotin MicroBeads for depletion. Thereby, highly pure CD4+ T cells are obtained.
Downstream applications Untouched CD4+ T cells may also be used for the isolation of specific T helper cell subsets, such as naive1 or memory CD4+ T cells and activated CD4+ T cells. |
