Detailed separation procedure For magnetic depletion of memory CD4+ T cells and non-CD4+ T cells, PBMCs are incubated with a cocktail of biotinylated CD45RO, CD8, CD14, CD15, CD16, CD19, CD25, CD34, CD36, CD56, CD123, anti-TCRγ/δ, anti-HLA-DR, and CD235a (glycophorin A) antibodies. These cells are subsequently magnetically labeled with Anti-Biotin MicroBeads for depletion.
Downstream applications Naive CD4+ T cells isolated with the Naive CD4+ T Cell Isolation Kit II can be used when direct labeling of cell surface molecules could interfere with downstream applications, e.g., in studies on naive T helper cell activation and in functional studies of DCs.1, 2 Untouched naive CD4+ T cells can also be used for further isolation of specific T helper cell subsets. For isolation of naive CD4+ recent thymic emigrants the CD4+ Recent Thymic Emigrant Isolation Kit, human is recommended. |
