From protein regulation to diseases The Nobel Prize-honored ubiquitin-proteasome system (UPS) establishes the major pathway for controlled protein degradation: It recognizes and destructs ubiquitin-tagged proteins. The UPS affects numerous processes, including cell cycle, DNA repair, and signal transduction. Any deregulation of this system – and its protein quality control – is associated with diseases, ranging from cancer to neurodegeneration.
Investigate UPS components and novel candidate genes! Disease-related changes within the UPS can be monitored and the impact of drug treatment on UPS components analyzed with the PIQORâ„¢ Ubiquitin-PS Microarrays (see fig. 1). Also, the tissue-specificity of the UPS components can be characterized.
Various microarray applications The PIQOR Ubiquitin-PS Microarray contains about 1,250 genes and allows the detection of mRNA expression changes along the entire ubiquitin-proteasome pathway. In addition, it contains a large number of unpublished candidate genes, identified using homology-based profile techniques, protein interaction analyses, and in cooperation with distinguished experts1-7.

Gene expression analysis of three human cell lines treated with the proteasomal inhibitor MG132 using PIQOR™ Ubiquitin-PS Microarrays. Jurkat, Raji and HEK293 cells were harvested after 1 h, 4 h and 24 h, respectively. Cluster analysis reveals up-regulated 26S-proteasomal subunits, relevant for the degradation of misfolded proteins (19S regulatory particle and β-subunits). The βi-subunits and the subunits of the PA28 regulator, specific to the immunoproteasome, are down-regulated. |
