The MACSelect™ Kk System uses the truncated mouse MHC class I molecule H-2Kk as a selection marker. H-2Kk expression is restricted to some rarely used mouse strains, e.g. AKR/J or CBA/J. MACSelect Kk can be used in virtually any mammalian cell line and in primary cells. The H-2Kk marker should not be used when MHC class I expression is down-regulated either directly or via reduced expression of β2 microglobulin, e.g. in some tumor cell lines such as adenocarcinoma cell lines. The pMACS Kk.II vector encodes the truncated H-2Kk surface marker and can be used for cloning the gene-of-interest. Additionally, it can be used for cotransfection in combination with a gene-of-interest expression vector.
The MACSelect Kk Transfected Cell Selection Kit provides the vectors, MACSelect MicroBeads, and fluorochrome-conjugated antibodies, including control vectors and antibodies required to set up and establish the MACSelect System. All kit components are also available separately. |
