| Overview |
The MSC Phenotyping Kit was developed for the standardized identification and phenotyping of cultured human MSCs by flow cytometry based on the defined ISCT standards1.
The MSC Phenotyping Kit includes reagents for:
- Enumeration of MSCs based on the expression of CD73, CD90, CD105
- Exclusion based on CD14, CD20, CD34, and CD45
- Control staining with the isotype cocktail
- Compensation with single color fluorochrome-conjugated antibodies: CD90, CD73, CD105.

Background information Mesenchymal stromal cells (MSCs) hold great potential for therapeutic applications. MSCs can be isolated from various tissues and expanded under different conditions. The cells are characterized by numerous phenotypic properties. Minimal criteria for a common definition of human MSCs was therefore defined by the Mesenchymal and Tissue Stem Cell Committee of the International Society for Cellular Therapy (ISCT) to facilitate comparability of data amongst investigators, accelerate new scientific discoveries, and support the development of novel cellular therapies1. The committee proposed that MSCs should be characterized by 1) plastic-adherence when maintained under standard culture conditions; 2) ability to differentiate into osteoblasts, adipocytes, and chondrocytes in vitro; 3) after culture, MSCs stain positive for CD73, CD90, and CD105 and negative for CD45, CD34, CD14 or CD11b, CD79α or CD19, and HLA-DR surface antigens1. |

| Ccultured MSCs was stained with the MSC Phenotyping Cocktail (purple) or with the Isotype Control Cocktail (black). The fractions were analyzed by flow cytometry using the MACSQuant® Analyzer. Please reference the datasheet for more details on the gating strategy. |
