| Overview |
| This antibody allows the direct identification of CD304 (BDCA-4/Neuropilin-1)+ plasmacytoid dendritic cells in blood and bone marrow by immunofluorescent staining and flow cytometric analysis.1,4 The antibody is suitable for staining of fresh or formaldehyde-fixed cells in suspension. Staining with CD304 (BDCA-4) antibodies is recommended for flow cytometric evaluation if plasmacytoid dendritic cells are isolated using the CD303 (BDCA-2) MicroBead Kit. |

Background information Clone AD5-17F6 recognizes the human CD304 (BDCA-4/Neuropilin-1) antigen. Like CD303 (BDCA-2), the CD304 (BDCA-4/Neuropilin-1) antigen1 is specifically expressed on human plasmacytoid dendritic cells in blood1–3 and bone marrow4. Unlike CD303 (BDCA-2), expression of CD304 (BDCA-4/Neuropilin-1) is up-regulated on plasmacytoid dendritic cells and induced on myeloid cells upon culturing.4 In inflamed tonsils, CD304 (BDCA-4/Neuropilin-1) expression, apart from plasmacytoid dendritic cells, is also detected primarily on follicular B helper memory T cells.4 CD304 (BDCA-4) is identical to neuropilin-1 (NP-1),4 a neuronal receptor, which is known to be expressed on numerous nonhematopoietic cell types, including neurons, endothelial, and tumor cells. CD1a+ dendritic cells generated ex vivo from monocytes or hematopoietic precursor cells are CD304 (BDCA-4/Neuropilin-1)+ but CD303 (BDCA-2)–.1 |
