Stem cells require special conditions for survival and growth and an important aspect of this is the underlying matrix. The reconstituted basement membrane (RBM) isolated from the mouse Engelbreth-Holm-Swarm (EHS) tumor is a matrix composed of laminin, collagen IV, heparin sulfate, proteoglycan, nidogen, entactin, and growth factors that supports the attachment, growth, and differentiation of many adherent cell types. RBM is able to support the growth and maintenance of mouse and human embryonic stem (ES) cells and induced pluripotent stem (iPS) cells.
Laminin, a glycosylated protein, is the main component of RBM. Laminin can serve as an alternative to mouse embryonic fibroblast (MEF) feeder cells for propagating pluripotent human or mouse cell lines and can be used to support derivation and differentiation of lineage-restricted progenitor cells. Also, pure laminin can support ES and iPS cell growth in the undifferentiated state, when it is desirable to have defined growth conditions. Stemgent® Laminin should be used as a cell attachment matrix when growth factors and other contaminants present in less pure RBM gels might compromise an experiment. |

| Stemgent’s primary stem cell lines have been successfully used in many laboratories for use as experimental controls or for studying epigenetic modifications. Mouse primary iPS cells-T1b and -WP5 are induced pluripotent stem (iPS) cells that have not been genetically modified. When cultured under standard mouse ES cell conditions, they are morphologically identical to mouse ES cells. They also express the pluripotency marker SSEA-1 and have strong AP activity. Mouse primary iPS cells-NNeo harbor the reverse tetracycline transactivator (rtTA), Oct4 cDNA, a neomycin resistance gene, and the doxycycline-inducible transcription factors Klf4, Sox2 and c-Myc. |
