Treg Suppression Inspector, human
Description The Treg Suppression Inspector has been developed for the functional characterization of human CD4+CD25+ regulatory T (Treg) cells by in vitro suppression assays. Treg cells are a subset of T cells that have the ability to suppress harmful immunological reactions to self or foreign antigens. This function of Treg cells can be analyzed by a so-called suppression assay. For this purpose, Treg cells and responder T cells are cocultured in different ratios in the presence of a polyclonal stimulus, in this case the Treg Suppression Inspector, while the proliferative response is measured. The Treg Suppression Inspector is an optimized T cell stimulation reagent for a Treg suppression assay, which is based on Anti-Biotin MACSiBeadâ„¢ Particles that are loaded with biotinylated CD2, CD3, and CD28 antibodies. Culture of responder T cells (CD4+CD25- or CD4+ T cells) alone in the presence of the Treg Suppression Inspector leads to proliferation of the responder T cells, whereas Treg cells alone show no significant proliferation. Coculture of Treg cells with responder T cells results in reduced proliferation of responder T cells due to the suppressive function of Treg cells. Applications The Treg Suppression Inspector is designed for the functional characterization of human CD4+CD25+ Treg cells by in vitro suppression assays.		Figure 1 CD4+CD25+ regulatory T cells, isolated with the CD4+CD25+ Regulatory T Cell Isolation Kit, were cocultured with CD4+CD25- responder T cells in different ratios. For T cell stimulation, the Treg Suppression Inspector was added to the culture. As controls, CD4+CD25+ Treg cells and CD4+CD25- responder T cells alone were cultured without any stimulus.
