High quality RNA or DNA samples
Accurate microarray analysis depends on preparation methods that circumvent common pitfalls, such as degradation or contamination of RNA and DNA. If you want to save time and resources and do not perform nucleic extraction methods routinely, then use our isolation service for RNA and DNA and benefit from reproducible microarray analyses. State-of-the-art quality control methods are used for quantification and qualification of RNA and DNA Alternatively, you can also provide us with total RNA samples for gene expression profiling or microRNA expression profiling analysis and with genomic DNA samples for array-CGH analysis. For the RNA-extraction from cells column-based methods are recommended while the well established Trizol protocol is the recommendation for cryoconserved tissue samples. For microRNA expression analysis it is essential to choose an RNA isolation method which preserves small RNA species. High quality intact DNA samples obtained with Qiagen or Gentra kits are crucial to the success of Agilent array-CGH experiments. Our service goes beyond simply assessing the quality and quantity of RNA and DNA samples. If necessary we can offer a variety of solutions to improve the RNA and DNA quality, such as DNAse treatment of RNA samples, proteinase treatment of DNA samples and re-purification. To ensure high quality starting material, we provide appropriate recommendations and detailed guidelines for RNA and DNA sample preparation and submission.

Various approaches for blood samples
Microarray analysis from blood samples involves various approaches for this demanding sample source, which comprises fresh or stabilized whole blood samples, blood fractions (e.g. PBMCs, serum, plasma) and separated blood cells. Choosing the right starting material is crucial to meaningful and reliable microarray results. The experienced Genomic Service Team can offer a variety of recommendations and solutions to meet this demand. Non-stabilized whole blood samples (EDTA, citrate) are only suitable for array-CGH analysis. Their usage for gene expression profiling should be avoided since the samples have to be processed immediately and the expression profile may change during short storage. Therefore, whole blood stabilization procedures, such as the standardized PAXgene™ Blood tubes (Qiagen) or Tempus™ Blood Tubes (Applied Biosystems), have to be used.
Miltenyi Biotec has developed numerous state-of-the-art tools for magnetic cell separation directly from whole blood Microarray analysis of whole blood samples involve the risk that differences in blood cell composition interfere with the expression profile. Gene expression analysis of isolated blood cells is highly recommended in order to circumvent this problem. Highly enriched and pure cell fractions can be obtained by using MACS Whole Blood Microbeads, tools for the gentle isolation of numerous leukocytes subsets. This reliable cell separation procedure can easily be standardized and used in an automated fashion with the autoMACS® Pro Separator. This synergy of MACS Technology and genomic analysis is called MACS-4-omics. Even rare cell populations resulting from blood cell separation can be further analyzed by using the unique SuperAmp™ Service.

From cell lines to primary cells
With ten years of microarray-experience, the Genomic Service Team ensures optimal processing of cell samples by applying well-proven protocols to cell lines or cells sorted by flow cytometry or MACS® Technology. MACS® Magnetic MicroBeads are perfect companions for the downstream gene expression analysis. The fast and easy cell separation procedure is pivotal for highly reproducible and accurate gene expression profiling. This synergy of MACS technology and genomic analysis is called MACS-4-omics. Even in cases where only small numbers of cells are obtained by cell sorting, it is still possible to perform reliable microarray analysis with the SuperAmp™ Service in combination with the Agilent Whole Genome Microarray Platform.
SuperAmp™ Technology is also the basic prerequisite for microarray analysis of cells obtained by LCM (Laser Capture Microdissection), a demanding starting material due to the low number of selected cells. The Genomic Services Team has used SuperAmp Technology for numerous LCM samples and is aware of the pitfalls during the sample collection.

Broad range of tissue sample preparation skills
The outcome of a microarray experiment depends on the quality of sample preparation. The experienced Genomic Service Team possesses a broad range of tissue sample preparation skills and uses well-established methods for different sample sources, ranging from cryoconserved tissues to tissue biopsies, from LCM (Laser Capture Microdissection) samples to particularly demanding FFPE (formalin-fixed-paraffin embedded) tissues. The isolation of total RNA from cryoconserved tissues requires a fast and thorough homogenization of the starting material. With the gentleMACS™ Dissociator Miltenyi Biotec has developed a powerful tool that meets this requirement with optimized homogenization protocols. The SuperAmp™ Service enables the performance of reliable microarray analysis from tissue sources with limited cell numbers, such as tissue biopsies and LCM samples from tissue sections. Based on their experience with LCM samples, the Genomic Services Team is aware of the pitfalls of sample collection and can provide qualified recommendations which are summarized in our guidelines for tissue sample preparation and submission. Archived biopsies are a precious source of information on molecular causes of various diseases and the sequelae of disease at a molecular level. The Microarray Services portfolio includes preparation of the particularly challenging FFPE (formalin-fixed-paraffin embedded) tissue samples. Established methods are used to ensure successful microarray experiments for gene expression profiling and array-CGH analysis.
