NEW: Anti-iNKT MicroBeads for the isolation of human invariant natural killer T cells expressing the TCR Vα24-Jα18.

Anti-iNKT MicroBeads have been developed for the separation of human invariant natural killer T cells (iNKT cells). This cell type is characterized by expression of an invariant T cell receptor α-chain and certain TCR β-chains (Vα24- Jα18 combined with Vβ11). iNKT cells represent a distinct lymphocyte population that co-expresses T cell and NK cell surface markers. iNKT cells are involved in immunoregulatory processes such as tolerance, host defense, and tumor surveillance.

NEW: Kit for the isolation of CD4+ Recent Thymic Emigrants

The CD4+ Recent Thymic Emigrant Isolation Kit is designed for the isolation of CD4+CD45RA+CD31+ recent thymic emigrants (RTE) from human PBMCs.<br>
The isolation is performed in a two-step procedure. First, non-CD4+ and memory CD4+ T cells are indirectly magnetically labeled with a cocktail of biotinylated antibodies and Anti-Biotin MicroBeads. The labeled cells are subsequently depleted over a MACS® Column. In the second step, CD4+CD45RA+CD31+ RTEs are directly labeled with CD31 MicroBeads and isolated by positive selection from the pre-enriched naive CD4+ T cell fraction.<br>
RTEs are a subpopulation of naive T cells characterized by their content of T cell receptor excision circles (TRECs).<br>
CD31 (platelet endothelial cell adhesion molecule-1, PECAM-1) has been described as a marker to identify the CD4+ RTEs among the naive CD4+ T cell population, as its expression correlates with the TREC content of CD4+ T cells. CD4+ T cells lacking CD31 expression harbor little or no TRECs, suggesting that they already proliferated in the periphery.<br>
Therefore, the magnetic isolation of CD4+ RTEs is possible by direct isolation of CD31+ cells from pre-enriched, untouched naive CD4+ T cells.

NEW: Kit for the isolation of naive human CD4+ T cells

The Naive CD4+ T Cell Isolation Kit II provides a fast and efficient way to isolate untouched naive CD4+ T cells from human peripheral blood. It is the new generation of the Naive CD4+ T Cell Isolation Kit, for even higher purities of the isolated naive T helper cell population.<br>
For magnetic depletion of memory CD4+ T cells and non-CD4+ T cells, PBMCs are incubated with a cocktail of biotinylated CD45RO, CD8, CD14, CD15, CD16, CD19, CD25, CD34, CD36, CD56, CD123, anti-TCRγ/δ, anti-HLA-DR, and CD235a (glycophorin A) antibodies. These cells are subsequently magnetically labeled with Anti-Biotin MicroBeads for depletion.

NEW: Kit for the isolation of human CD4+CD25+CD45RA+ regulatory T cells

The CD4+CD25+CD45RA+ Regulatory T Cell Isolation Kit has been developed for the isolation of CD45RA+ regulatory T cells from human PBMCs. The isolation is performed in a two-step procedure. First, non-CD4+ and memory T cells are indirectly magnetically labeled with a cocktail of biotin-conjugated antibodies and Anti-Biotin MicroBeads. The labeled cells are subsequently depleted by separation over a MACS® Column.<br>
In the second step, CD4+CD25+CD45RA+ T cells are directly labeled with CD25 MicroBeads and isolated by positive selection from the pre-enriched CD4+CD45RA+ T cell fraction.<br>
Regulatory CD4+CD25+ T cells isolated from the CD45RA+ naive T cell compartment were shown to be optimal for in vitro expansion. These expanded regulatory T cells maintained the FoxP3+ phenotype and their suppressive activity, whereas CD45RA– regulatory T cells from the memory T cell compartment lost FoxP3 expression and showed only modest suppressive activity.

The PepTivator™ EBV BZLF1 is a peptide pool that consists mainly of 15-mer peptides with 11 amino acid (aa) overlap. It has been developed for efficient <i>in vitro</i> stimulation of EBV BZLF1–specific CD4+ and CD8+ T cells as peptides of 15-aa length with 11-aa overlap represent an optimized solution for stimulating both CD4+ and CD8+ T cells in various applications.<br>
Epstein-Barr virus (EBV) is a human γ-herpesvirus with B cell growth–transforming ability and lymphomagenic potential. More than 90% of human adults are infected with EBV. Usually the primary infection is asymptomatic, but some individuals develop an infectious mononucleosis, a self-limiting lymphoproliferative disease.<br>
BZLF1 activates a promoter of an EBV gene and plays a key role in the switch from latent to productive infection of EBV. BZLF1-specific CD4+ and CD8+ T cells have been found in EBV-infected individuals.

The Anti-MSCA-1 (W8B2) MicroBead Kit, based on the antibody clone W8B2, was developed for the isolation of mesenchymal stromal cells (MSCs) from bone marrow aspirates. The kit contains all necessary reagents for the magnetic isolation of MSCA-1+ cells, including Anti-MSCA-1 (W8B2) MicroBeads for magnetic labeling and FcR Blocking Reagent to block non-specific labeling via Fc receptors. The Anti-MSCA-1 (W8B2) MicroBeads recognize the mesenchymal stromal cell antigen-1 (MSCA-1), a so far unknown antigen. MSCA-1 was shown to be restricted to MSCs in the CD271bright population in bone marrow. These CD271brightCD45dim MSCs have a much higher clonogenic capacity compared to the CD271+CD45+ fraction in bone marrow. Therefore, MSCA-1 is a highly specific marker to isolate MSCs with a high proliferative potential from bone marrow. MSCA-1 expression was not found on placenta-derived MSCs after culture. For the optimal separation and subsequent expansion of MSCA-1+ cells, the MSC Research Tool Box – MSCA-1 (W8B2) is available.

NEW: CD146 MicroBead Kit, human--for isolation of mesenchymal stromal cells as well as endothelial cells and pericytes

The CD146 MicroBead Kit was developed for the isolation of CD146+ cells. The kit consists of CD146 MicroBeads and FcR Blocking Reagent. The CD146 MicroBeads recognize the human CD146 antigen, also known as MUC18, MCAM, Mel-CAM, and S-Endo-1. CD146 is a transmembrane glycoprotein belonging to the immunoglobulin superfamily and contains five extracellular immunoglobulin (Ig)-like domains.<br>
CD146 possesses a limited tissue expression distribution, including endothelial cells, smooth muscle cells, pericytes, follicular dendritic cells, melanoma cells, and a subpopulation of activated T lymphocytes. CD146+ cells isolated from umbilical cord, lipoaspirate, dental pulp, or endometrial tissue, are described to contain cells with clonogenic fibroblastic colony (CFU-F) activity and multilineage differentiation potential, which are often called mesenchymal or marrow stromal cells (MSCs).<br>
CD146 functions as a Ca2+-independent cell adhesion molecule, which is involved in heterophilic cell-cell interactions, and may be involved in the extravasion and/or homing of activated T cells. CD146 expression in melanoma is also directly associated with tumor growth and metastasis.<br>

Treg cells are a subset of T cells with the ability to suppress harmful immunological reactions to self and foreign antigens. Due to the lack of a specific marker for Treg cells, a combination of CD4, CD25, and FoxP3 is commonly used for their identification. The Treg Detection Kit includes all reagents necessary for cell surface staining of CD4 and CD25, and intracellular staining of FoxP3 with a special FoxP3 Staining Buffer Set.
NEW: Memory B Cell Isolation Kit

The Memory B Cell Isolation Kit has been developed for the isolation of memory B cells from human PBMCs. Memory B cells, defined as quiescent antigen-experienced B cells, are generated in response to T cell–dependent and T cell–independent antigens. They are able to react quickly to a recurrent antigenic challenge, thereby providing serological immune protection.<br>
CD27, a member of the TNF receptor family, is expressed on most memory B cells. It allows their discrimination from naive B cells, which are CD27–.<br>
Memory B cells are isolated by depletion of unwanted cells and subsequent positive selection with CD27 MicroBeads. Unwanted cells, for example, T cells, NK cells, monocytes, dendritic cells, granulocytes, platelets, and erythroid cells are depleted using a cocktail of biotinylated antibodies against CD2, CD14, CD16, CD36, CD43, and CD235a (glycophorin A), and Anti-Biotin MicroBeads. For evaluation of MACS® Separations, staining with CD19-APC and CD27-PE antibodies is recommended.

MICROGLIA: CD11b (Microglia) MicroBeads for the isolation of human microglial cells.

Microglia, often called brain macrophages, are the resident immune effector cells in the central nervous system (CNS). In addition, activated microglia serve as the major antigen-presenting cells in the CNS. They are morphologically, immunophenotypically, and functionally related to cells of the monocyte/macrophage lineage. The most characteristic feature of microglial cells is their rapid activation in response to injury, inflammation, neurodegeneration, infection, and brain tumors.<br>
A paucity of microglial antigens has hindered microglial identification. However, CD11b has been widely used as marker for microglial identification and is also suitable for their immunomagnetic isolation from human brain tissue.<br>

STIMULATING: Peptide pools for efficient in vitro stimulation of human CMV- and AdV-specific T cells

The PepTivator - CMV IE-1 is a peptide pool that consists mainly of 15-mer peptides with 11–amino acid (aa) overlap, covering the complete sequence of the IE-1 protein of human cytomegalovirus (CMV).<br>
The PepTivator – AdV5 Hexon is a peptide pool that consists mainly of 15-mer peptides with 11–amino acid overlap.<br>
Adenoviruses (AdV) are non-enveloped, ubiquitous, lytic double-stranded DNA viruses. Up to now, 52 different human serotypes (1–52), divided into six species (A-F) are described.<br>
The hexon protein is one of the structural proteins in the viral coat and is synthesized during late infection. It is the immunodominant T cell target among capsid proteins and contains multiple epitopes conserved among serotypes.<br>

HUMAN TREGS: CD39 antibodies for characterization of human regulatory T cells and other CD39-expressing cells

The CD39 antibody specifically recognizes the membrane-bound human CD39 antigen, which is expressed on an effector/memory-like subset of FoxP3+ regulatory T cells1. CD39 is an ectonucleotidase and catalyzes the hydrolysis of extracellular nucleotides, for example, ATP. In concert with CD73, which is an ecto-5’-nucleotidase, this can lead to the production of adenosine.<br>
High extracellular ATP concentrations indicate tissue injury and cell death and induce various pro-inflammatory responses in immune cells.<br>
Through its enzymatic activity, CD39 can contribute to the suppressive function of regulatory T cells, for example, by eliminating extracellular ATP or by generating adenosine, which has suppressive effects on various immune cells.

The Treg Suppression Inspector has been developed for the functional characterization of human CD4+CD25+ regulatory T (Treg) cells by in vitro suppression assays.<br>
Treg cells are a subset of T cells that have the ability to suppress harmful immunological reactions to self or foreign antigens. This function of Treg cells can be analyzed by a so-called suppression assay. For this purpose, Treg cells and responder T cells are cocultured in different ratios in the presence of a polyclonal stimulus, in this case the Treg Suppression Inspector, while the proliferative response is measured.

CHILLING: MACS Cooling Tube Racks

MACS® Cooling Tube Racks are valuable tools for any procedure where cooling of sample tubes is desired. Cooling of cell samples helps to sustain cell viability during cell analysis or cell separation. The tube racks are pre-cooled in the refrigerator (2–8 °C). The special coolant contained in the tube racks switches from fluid to solid and maintains a low temperature until the coolant is completely melted.

HUMAN NEUTROPHILS: Separation and analysis using CD66abce MicroBead Kit and Antibodies

CD66abce MicroBeads are used for the isolation or depletion of human neutrophils from lysed human peripheral blood. Neutrophils play a prominent role in the innate immune response through their phagocytic activity. They are abundant in peripheral blood but almost absent in healthy tissue. Upon infection, large numbers of neutrophils are recruited to the sites of inflammation.<br>
The CD66abce Antibody is suitable for the identification and enumeration of CD66abce+ human cells by flow cytometry or fluorescence microscopy.

GENTLE AND EFFICIENT: Yield viable neuronal progenitor cells of high purity within just hours with the Neural Tissue Dissociation Kit (Trypsin) and Anti-PSA-NCAM MicroBeads.

The Neural Tissue Dissociation Kit (Trypsin) has been developed for the gentle, but efficient, generation of single-cell suspensions from neural tissues. In less than an hour, the gentle two-step enzymatic dissociation procedure efficiently yields high numbers of viable cells. Neural tissues from embryonic, postnatal and adult murine tissue sections or whole brain preparations can be processed. Dissociated cells are immediately ready for further in vitro or in vivo applications, including MACS® Cell Separations.<br>
Anti-PSA-NCAM MicroBeads have been developed for the isolation of PSA-NCAM+ cells. The anti-PSA-NCAM antibody recognizes polysialic acid (PSA) which, in vertebrates, is linked to the extracellular domain of the neural cell adhesion molecule (NCAM, CD56). PSA-NCAM, the highly polysialated form of NCAM, is predominantly expressed in embryonic and neonatal neural tissue. In adult mammalian brain PSA-NCAM expression is restricted mainly to areas that retain neurogenic potential, such as the subventricular zone (SVZ) and the dentate gyrus of the hippocampus.

EVEN BETTER: CD4+CD62L+ T Cell Isolation Kit II for the isolation of naive mouse CD4+ T cells – new version!

The CD4+CD62L+ T Cell Isolation Kit II has been developed for the isolation of CD4+CD62L+ T helper cells from spleen and lymph nodes.<br>
CD62L (L-selectin) is highly expressed on naive T cells and down-regulated upon activation. It is also expressed on a small subset of memory T helper cells, the central memory T cells, which can be distinguished from naive T helper cells by their high expression of CD44. Furthermore, CD62L is expressed on most thymocytes, naive CD8+ T cells, B cells, dendritic cells, macrophages, NK cells, neutrophils, eosinophils, regulatory T cells, and TCRγ/δ+ T cells. For isolation of CD4+CD62L+ T helper cells, the non-T helper cells as well as regulatory T cells and γ/δ T cells are depleted by indirect magnetic labeling using a cocktail of lineage-specific biotin-conjugated antibodies against CD8a (Ly-2), CD45R (B220), CD49b (DX5), CD11b (Mac-1), and Ter-119, as well as antibodies against CD25 and TCRγ/δ in combination with Anti-Biotin MicroBeads. Subsequently, CD4+CD62L+ T cells are positively selected from the enriched CD4+ T helper cell fraction with CD62L (L-selectin) MicroBeads.

NEW: Products for the enrichment of activated mouse CD154+ T helper cells.

The CD154 Enrichment and Detection Kit is designed for the easy identification and enrichment of antigen-specific CD4+ T cells. Detection and stimulation of the cells are conveniently performed in one step.<br>
CD154, also known as CD40L, gp39, T-BAM, and TRAP is transiently up-regulated on activated CD4+ T cells and plays an important role as a costimulatory molecule in the interaction between T cells and antigen-presenting cells through ligation of CD40. Due to its transient expression within hours of activation, CD154 can be used as a marker for activated antigen-specific CD4+ T cells.<br>
Using the CD154 Enrichment and Detection Kit, antigen-specific CD4+ T cells are indirectly magnetically labeled with a CD154 Detection Cocktail (PE) and anti-PE MicroBeads.<br>
To optimize the detection of CD154 on stimulated antigen-specific CD4+ T cells, cells are incubated simultaneously with the desired antigen in vitro to induce the expression of CD154 and with the CD154 Detection Cocktail. Besides a PE-conjugated CD154 antibody, the CD154 Detection Cocktail contains a CD40 blocking antibody to prevent the down-regulation of CD154 surface expression, and a CD28 antibody to provide a costimulatory signal. <br>
After stimulation, cells are magnetically labeled with anti-PE MicroBeads and separated on a MACS Column.

USEFUL COMPANION: A special protocol for the isolation of mouse CD105+Sca-1+ LTR-HSCs is now available.

This special protocol describes the isolation of CD105+ Sca-1+ HSCs from mouse bone marrow using a two-step magnetic purification procedure. After initial selection for CD105 expression, purified cells are magnetically labeled for a second time to select for Sca-1 expression. After the second step, the resulting cell population will be CD105+ Sca-1+ LTR-HSCs.<br>
Sequential magnetic purification of the target cells is facilitated by using the CD105 MultiSort Kit (PE), mouse (# 130-092-924) in the first dimension, which includes a MicroBead removal step before cells are labeled for a second time with the Anti-Sca-1 MicroBead Kit (FITC), mouse (# 130-092-529).<br>

FoxP3, also known as FORKHEAD BOX P3, SCURFIN, and JM2, is a member of the forkhead/winged-helix family of transcriptional regulators. It is expressed predominantly in Tregs and is a major marker and functional regulator of T reg cell development and function.Mutations in the FoxP3 gene have been shown to be linked to the autoimmune manifestations observed in the Scurfy mouse and in humans with immune dysregulation, polyendocrinopathy, enteropathy, X-linked syndrome (IPEX) disease. Studies in the mouse showed that FoxP3-deficient animals lack T reg cells, whereas overexpression of the FoxP3 protein leads to profound immune suppression1. The Anti-FoxP3 antibody, clone 3G3, detects human as well as mouse FoxP3 2. Read more on our website.

NEW: Intracellular Cytokine Detection Kits

Miltenyi Biotecs Intracellular Cytokine Detection Kits are designed for rapid and reliable flow cytometric detection of cytokine production in activated CD4+ T cells. As a specific and functional marker of T cell activation, CD154 is used. CD154 is transiently up-regulated on activated CD4+ T cells within hours after activation. By combining the detection of cytokine secreting cells with staining of CD154 and CD4, antigen specific TH cell responses can be directly identified. <br>
The Kits contain all reagents necessary for the detection of cytokine-producing activated CD4+ T cells from whole blood or PBMCs by intracellular staining. In addition, reagents are included for the fixation and permeabilization of cells prior to immunofluorescent staining, as well as for the in vitro stimulation of T cells as a positive control.
